J&J and Merck Ebola vaccines produce lasting antibodies in children and adults

Ebola vaccines developed by Johnson & Johnson and Merck & Co produced virus-fighting antibodies and appear to be safe in children and adults, according to data from two studies published on Wednesday.

Both companies’ vaccines produced antibodies 14 days after the first of two shots and were detectable at varying levels in both children and adults for one year, data from the studies conducted in Western Africa showed.

The vaccines are designed to target the Zaire strain of the virus, not the Sudan strain of Ebola that recently caused an outbreak and at least 56 deaths in Uganda.

One regimen tested a dose of J&J’s vaccine, followed by a booster shot of a vaccine from Danish drugmaker Bavarian Nordic, while another tested two doses of Merck’s vaccine with eight weeks in between. A third option followed the first Merck dose with a placebo.

“I think the study shows that both the vaccines elicit good antibody responses,” said Dr. H. Clifford Lane, one of the researchers and a clinical director at the U.S. National Institute of Allergy and Infectious Diseases, part of the National Institutes of Health (NIH).

Only Merck’s shot can potentially be given as a single dose while J&J’s vaccine may need to continue as a two-dose regimen, Lane added.

The NIH researchers noted that they were unable to assess the actual level of protection against the disease from the vaccines as no participants contracted Ebola during the trial, which began enrollment in 2017. But they said the vaccines were found to be safe for children and adults.

“Long-term follow up of the participants in this trial is taking place to determine if and when booster doses might be needed,” said Brian Greenwood, a study co-author from the London School of Hygiene & Tropical Medicine.

A total of 1,400 adults and 1,401 children aged 1 to 17 years old participated in the trials conducted in collaboration with Liberia Ministry of Health and with the University Clinical Research Center and the Center for Vaccine Development-Mali.

The results were published in The New England Journal of Medicine.

Merck’s vaccine Ervebo was approved by the European regulator and prequalified by the World Health Organization in 2019. J&J’s Zabdeno got European and WHO clearances in 2020 and 2021, respectively. Bavarian Nordic’s Mvabea, used in the J&J regimen, also received European approval in 2020 and WHO prequalification in 2021.

We Finally Know How Black Holes Produce The Most Brilliant Light in The Universe : ScienceAlert

For something that emits no light that we can detect, black holes just love to cloak themselves in radiance.

Some of the brightest light in the Universe comes from supermassive black holes, in fact. Well, not actually the black holes themselves; it’s the material around them as they actively slurp down vast amounts of matter from their immediate surroundings.

Among the brightest of these maelstroms of swirling hot material are galaxies known as blazars. Not only do they glow with the heat of a swirling coat, but they also channel material into ‘blazing’ beams that zoom through the cosmos, shedding electromagnetic radiation at energies that are hard to fathom.

Scientists have finally figured out the mechanism producing the incredible high-energy light that reaches us from billions of years ago: Shocks in the black hole’s jets that boost the speed of particles to mind-blowing velocities.

“This is a 40-year-old mystery that we’ve solved,” says astronomer Yannis Liodakis of the Finnish Centre for Astronomy with ESO (FINCA). “We finally had all of the pieces of the puzzle, and the picture they made was clear.”

Most of the galaxies in the Universe are built around a supermassive black hole. These mind-bogglingly large objects sit in the galactic center, sometimes doing very little (like Sagittarius A*, the black hole at the heart of the Milky Way) and sometimes doing a lot.

That activity consists of accreting material. A vast cloud assembles into an equatorial disk around the black hole, circling it like water around a drain. The frictional and gravitational interactions at play in the extreme space surrounding a black hole cause this material to heat up and shine brightly across a range of wavelengths. That’s one source of a black hole’s light.

The other – the one at play in blazars – are twin jets of material launched from the polar regions outside the black hole, perpendicular to the disk. These jets are thought to be material from the inner rim of the disk that, rather than falling toward the black hole, gets accelerated along external magnetic field lines to the poles, where it is launched at very high speeds, close to the speed of light.

For a galaxy to be classified as a blazar, these jets have to be almost directly pointed toward the viewer. That’s us, on Earth. Thanks to extreme particle acceleration, they blaze with light across the electromagnetic spectrum, including high-energy gamma- and X-rays.

Exactly how this jet accelerates the particles to such high speeds has been a giant cosmic question mark for decades. But now, a powerful new X-ray telescope called the Imaging X-ray Polarimetry Explorer (IXPE), launched in December 2021, gave scientists the key to solve the mystery. It’s the first space telescope that reveals the orientation, or polarization, of X-rays.

“The first X-ray polarization measurements of this class of sources allowed, for the first time, a direct comparison with the models developed from observing other frequencies of light, from radio to very high-energy gamma rays,” says astronomer Immacolata Donnarumma of the Italian Space Agency.

IXPE was turned to the brightest high-energy object in our sky, a blazar called Markarian 501, located 460 million light-years away in the constellation of Hercules. For a total of six days in March 2022, the telescope collected data on the X-ray light emitted by the blazar’s jet.

At the same time, other observatories were measuring the light from other wavelength ranges, from radio to optical, which previously were the only data available for Markarian 501.

The team soon noticed a curious difference in the X-ray light. Its orientation was significantly more twisted, or polarized, than the lower-energy wavelengths. And the optical light was more polarized than the radio frequencies.

However, the direction of the polarization was the same for all wavelengths and aligned with the direction of the jet. This, the team found, is consistent with models in which shocks in the jets produce shockwaves that provide additional acceleration along the length of the jet. Closest to the shock, this acceleration is at its highest, producing X-radiation. Farther along the jet, the particles lose energy, producing lower-energy optical and then radio emission, with lower polarization.

“As the shock wave crosses the region, the magnetic field gets stronger, and energy of particles gets higher,” says astronomer Alan Marscher of Boston University. “The energy comes from the motion energy of the material making the shock wave.”

It’s not clear what creates the shocks, but one possible mechanism is faster material in the jet catching up to slower-moving clumps, resulting in collisions. Future research could help confirm this hypothesis.

Since blazars are among the most powerful particle accelerators in the Universe, and one of the best laboratories for understanding extreme physics, this research marks a pretty important piece of the puzzle.

Future research will continue to observe Markarian 501, and turn IXPE to other blazars to see if similar polarization can be detected.

Rockstar To Let Players Produce Their Own Beats for GTA Online

When you hear of Rockstar Games and music, it’s almost like the latter is a synonym of the former. Because music is simply part of Rockstar’s DNA. Sam Houser, the founder of Rockstar Games, is incredibly passionate about anything music-related. Just last year, Rockstar launched a record label in partnership with CircoLoco.

This strong inspiration for music artistry is materializing more evidently within Grand Theft Auto Online in the past two years. Starting with The Cayo Perico Heist update in December 2020, which featured Moodymann, Joy Orbison, Palms Trax, Keinemusik, and cameo appearances of West Coast legends Dr. Dre and DJ Pooh.

Rockstar is not stopping any time soon to feature more music-related content. Within the game files of the “Contract” update, Dr. Dre’s VIP Contract is referred to as the first VIP Contract. Suggesting a second VIP Contract may arrive in the future. Plus, a bunch of new, unused for now, parameters were added, within the “Criminal Enterprises” update, to functions associated with the DJ Tracks.

But more importantly, Rockstar’s next grand idea for music? As per a reliable source, Rockstar will let players come up with their own mixes and beats in Grand Theft Auto Online. Similarly, an interesting development occurred with how Take-Two Interactive, Rockstar’s parent company, registered a domain by the name of “rapponator.com” back in October 2020. A decade ago, in September 2009, Rockstar released a music mixer by the name of Beaterator in collaboration with Timbaland, allowing the player to produce their own loops. “The two names surely bear interesting resemblance as Rockstar likes to make references to previous product releases.

This idea has been stewing within Rockstar’s oven for a while. As well as Rockstar considering featuring Michael De Santa for the first time in GTA Online. Just like how half of the Contract update focused on Franklin Clinton and the other half focused on Dr. Dre. We could see a future update with one half focusing on Michael and the other half on making up your own mixes/beats.

Fire breaks out at world’s biggest produce market in Paris

PARIS — A billowing column of dark smoke towered over Paris on Sunday from a warehouse blaze at a massive produce market that supplies the French capital and surrounding region with much of its fresh food and bills itself as the largest of its kind in the world.

Firefighters urged people to stay away from the area in Paris’ southern suburbs, as 100 officers and 30 fire engines battled the blaze at the Rungis International Market.

Capt. Marc Le Moine, a spokesman for the Paris fire service, said no one was injured. The fire was brought under control and there was no risk of it spreading from the soccer field-sized warehouse, covering an area of 7,000 square meters (1.7 acres), he said.

The cause of the blaze was unknown but will be investigated, he added.

The sprawling wholesale market is a veritable town unto itself, with more than 12,000 people working there and warehouses filled with fruit and vegetables, seafood, meats, dairy products and flowers from across France and around the world.
