J&J vaccine adds to COVID-19 armoury, includes South African variant

(Reuters) – Johnson & Johnson said on Friday that its single-dose vaccine was 66% effective in preventing COVID-19 in a large global trial against multiple variants, giving health officials another weapon to tackle the pandemic.

In the trial of nearly 44,000 volunteers, the level of protection against moderate and severe COVID-19 varied from 72% in the United States, to 66% in Latin America and just 57% in South Africa, from where a worrying variant has spread.

The data showed that the vaccine’s effect on the South Africa variant was diminished compared to the unaltered virus, but infectious disease and public health experts said it can still help contain the virus spread and prevent deaths.

Midstage trial data from Novavax on Thursday also documented lower effectiveness in South Africa.

Rival shots from Pfizer/BioNTech and Moderna were both around 95% effective in preventing symptomatic illness in pivotal trials when given in two doses.

Those trials were conducted mainly in the United States and before the emergence of new variants. These mean that the world is racing against time and with limited supplies to vaccinate as many people as possible, and quickly, to prevent virus surges.

COVID-19 is rising in 37 countries and infections have surpassed 101 million globally.

Top U.S. infectious disease specialist Anthony Fauci said the world needs to vaccinate quickly to try to get ahead of these changes in the virus.

“It’s really a wake up call for us to be nimble and to be able to adjust as this virus will continue for certain to evolve,” Fauci said.

J&J’s main goal was the prevention of moderate to severe COVID-19, and the vaccine was 85% effective in stopping severe disease and preventing hospitalization across all geographies and against multiple variants 28 days after immunization.

That “will potentially protect hundreds of millions of people from serious and fatal outcomes of COVID-19,” Paul Stoffels, J&J’s chief scientific officer, said.

J&J shares were down 4% at $162.7 at 1700 GMT, with some Wall Street analysts saying its vaccine’s effectiveness was below those of rivals. Moderna’s stock gained 8% to $172.80.

SEEKING APPROVAL

J&J plans to seek emergency use authorization from the U.S. Food and Drug Administration next week and will soon follow up with the European Union and the rest of the world.

It has said it plans to deliver 1 billion doses of the vaccine, which it will make in the United States, Europe, South Africa and India, in 2021.

Public health officials are counting on it to increase much-needed supply and simplify immunization in the United States, which has a deal to buy 100 million doses of J&J’s vaccine and an option for an additional 200 million.

J&J said the vaccine would be ready immediately upon emergency approval, but Stoffels declined to say how many doses.

“The key is not only overall efficacy but specifically efficacy against severe disease, hospitalization, and death,” said Walid Gellad, a health policy associate professor at the University of Pittsburgh.

J&J’s vaccine uses a common cold virus to introduce coronavirus proteins into cells and trigger an immune response, whereas the Pfizer/BioNTech and Moderna vaccines employ a new technology called messenger RNA.

Unlike these vaccines, J&J’s does not require a second shot weeks after the first or need to be kept frozen, making it a strong candidate for use in parts of the world where transportation and cold storage present problems.

“Most countries are still desperate to get their hands on doses, regardless of whether or not the vaccine is considered highly effective. Moderately effective will do just fine for now,” Michael Breen, Director of Infectious Diseases and Ophthalmology at research firm GlobalData, said.

‘OVERWHELMED’

Several studies have emerged this month showing that a South African variant has mutated in areas of the virus that are key targets of vaccines, reducing their efficacy.

“What we are learning is there is different efficacy in different parts of the world,” Stoffels told Reuters.

In a sub-study of 6,000 volunteers in South Africa, Stoffels said, the J&J vaccine was 89% effective at preventing severe disease. In the South Africa portion of the trial, 95% of cases were infections with the South African variant.

“I am overwhelmed by the fact that this vaccine protected against severe disease even in South Africa,” said Glenda Gray, the joint lead investigator of the South African vaccine trial.

In the J&J trial, which was conducted in eight countries, 44% of participants were from the United States, 41% from Central and South America and 15% from South Africa. Just over a third of the volunteers were over 60.

Expert explains why he’d take the Johnson & Johnson vaccine

Johnson & Johnson’s single-shot Covid-19 vaccine was shown to be 66% effective in preventing moderate and severe disease in a global Phase 3 trial, but 85% effective against severe disease, the company announced Friday.

The vaccine was 72% effective against moderate and severe disease in the US, the company said.

It’s a striking difference from vaccines from Pfizer/BioNTech and Moderna, and it may give pause to people uncertain about which vaccine to get or when they can get one. The vaccines already on the market in the US are about 95% effective overall against symptomatic Covid-19, with perhaps even higher efficacy against severe cases.

Experts say the Johnson & Johnson vaccine, developed by its vaccine arm Janssen, will still be useful against the pandemic in the United States and around the world, even if the single-dose vaccine doesn’t become the first choice for many.

For Johnson & Johnson, efficacy against moderate and severe ranged from one country to another: 72% in the US, 66% in Latin America and 57% in South Africa. This was measured from starting one month after the shot.

In South Africa, 95% of cases in the trial were due to a variant known as B.1.351, which is known to be more contagious and carries mutations that may make the virus less susceptible to the antibody immune response – including antibodies prompted by vaccination.

With that variant, “we have a lower protection against milder forms of Covid than we did in the United States, where there were more typical circulating variants,” Dr. Mathai Mammen, the company’s global head of research and development, told CNN’s Chief Medical Correspondent, Dr. Sanjay Gupta.

Even those who got moderate cases of Covid-19 in the trial tended to develop a milder course and fewer symptoms, he added.

But for Mammen, the key result was how effective the vaccine was at preventing severe disease – regardless of variant or age group.

“Across all geographies, across all variants, we see 85% protection” against severe disease, he said. That trend increased over time, with no severe cases in the vaccinated group after day 49, according to the company.

From one month after the shot, all hospitalizations and deaths occurred in the placebo group.

The results are based on an analysis of more than 44,000 participants in eight countries, with 468 total cases of Covid-19 split between those receiving the vaccine or placebo. The results have not been published in a peer-reviewed journal, but the company said it plans to do so “in the coming weeks.”

Johnson & Johnson is expected to apply to the US Food and Drug Administration next week for emergency use authorization, which Mammen said could come by late February.

Report: Eagles hire Brian Johnson, the QB’s coach who helped Dak Prescott reach NFL

The Eagles are hiring University of Florida offensive coordinator Brian Johnson as their quarterbacks coach, according to NFL Network’s Ian Rapoport.

Johnson, 33, will replace Press Taylor, who will not be retained from Doug Pederson’s staff.

Johnson has been with the Gators since 2018. He joined them as quarterbacks coach and became offensive coordinator in the 2020 season.

Before then, Johnson spent one year in Houston as their OC/QBs coach and three years with Mississippi State before that. At Mississippi State, Johnson helped Dak Prescott become an NFL draft pick. Prescott has gone on to have great success in the NFL with the Dallas Cowboys, becoming the NFL Rookie of the Year in 2016. Prescott has been a two-time Pro Bowler and threw for nearly 5,000 yards in 2019. He was well on his way to passing that in 2020 before suffering a season-ending injury.

So now the Eagles will hope that the guy who helped create Dak will be able to help fix Carson Wentz.

The next big question is whether or not Wentz is back for the 2021 season. The relationship between Wentz and the Eagles is clearly in need of repair. But if Wentz stays in 2021, it will be Johnson who will be responsible for helping to fix Wentz.

But it’s also worth pointing out, as this Philadelphia Inquirer story from Mike Sielski notes, that Johnson recruited Jalen Hurts while at Mississippi State and has known Hurts since Hurts was 4 years old.

Johnson interviewed for head coaching jobs and South Carolina and Boise State this offseason and is considered one of the top young offensive coaches in the country.

It will be Johnson, along with head coach Nick Sirianni and offensive coordinator Shane Steichen in the top offensive coaching positions for the Eagles heading into next season. This will be an extremely young coaching staff. Sirianni is 39, passing game coordinator Kevin Patullo is 39, Steichen is 35, Johnson is 33 and defensive coordinator Jonathan Gannon is 37.

In addition to coaching Prescott, Johnson also helped Gators quarterback Kyle Trask put up some major numbers in 2020. Trask completed nearly 69% of his passes for 4,283 yards with 43 touchdowns and just 8 interceptions in 2020. Before Trask, Johnson coached up Feleipe Franks.

Johnson, who played quarterback at Utah, began his coaching career there in 2010 as the QBs coach. He was then promoted to offensive coordinator for two seasons before leaving for Mississippi State.

Boris Johnson urges new trade deal in first call with President Biden

British Prime Minister Boris Johnson urged President Biden to strike a new trade deal with the UK in a phone call on Saturday, marking the first official discussion between the two world leaders since Biden was sworn in.

Johnson “reiterated his intention to resolve existing trade issues as soon as possible” and discussed “the benefits of a potential free trade deal” with Biden, according to a statement from Downing Street reported by the Associated Press.

Johnson, a one-time Trump ally who distanced himself from the former president during his final term, also congratulated Biden on rejoining the Paris climate agreement and World Health Organization, two reversals of moves made by Trump.

“Great to speak to President @JoeBiden this evening,” Johnson tweeted Saturday.

“I look forward to deepening the longstanding alliance between our two countries as we drive a green and sustainable recovery from COVID-19.”

The Biden administration has stated in recent weeks that it is not ready to commit to any new trade deals.

“President Biden has been clear that he will not sign any new free trade agreements before the U.S. makes major investments in American workers and our infrastructure,” Treasury secretary nominee Janet Yellen said earlier this week.

The call with Johnson was at least Biden’s third with a foreign leader since taking office on Wednesday; the president spoke with Canadian Prime Minister Justin Trudeau and Mexican President Andres Manuel Lopez Obrador on Friday.

High-risk sports could resume in NY in February
High-risk sports can restart in New York State on Feb. 1 if local health departments approve. This includes basketball, wrestling, hockey, volleyball, football and lacrosse. However, whether or not they resume will rely on factors such as if there have been more cases of the more transmissible COVID variant, the local rates of COVID positivity and the ability to monitor and enforce compliance. Nassau County Executive Laura Curran says that schools can resume sports in the county.

2 more vaccine mega sites open in NJ
All six of the New Jersey’s COVID-19 vaccine mega sites are now open, with residents lining up Friday at two new locations in East Rutherford and Atlantic City. This as the state announced they have successfully vaccinated 500,222 residents in a little more than a month. Sites at the Meadowlands Racing and Entertainment Complex and Atlantic City Convention Center were both fully booked, but more appointment times are expected to be added based on availability.

UK chief scientist says new COVID-19 variant may be more deadly, more research needed
There is some evidence that a new coronavirus variant first identified in southeast England carries a higher risk of death than the original strain, the British government’s chief scientific adviser said Friday — though he stressed that the data is uncertain. Patrick Vallance told a news conference that “there is evidence that there is an increased risk for those who have the new variant.” He said that for a man in his 60s with the original version of the virus, “the average risk is that for 1,000 people who got infected, roughly 10 would be expected to unfortunately die.”

“With the new variant, for 1,000 people infected, roughly 13 or 14 people might be expected to die,” he said.

Researchers at UC San Diego’s School of Engineering are working on a potential game changer in the fight against COVID-19. Researchers are looking into a new type of test that could detect the virus on your face mask. The test can be worn as a sticker on your mask. The sticker includes a little dye and works just like an at-home pregnancy test. As someone wears the sticker on the mask throughout the day, it collects droplets. After a few hours, it can detect COVID-19 molecules from your breath.
